The shrine is located in the East section of map Campagna District right along the "dashed white line" just North of the Termi Di Diocleziano monument there is a tunnel entrance right next to the monument as well.
When you are able to gain access to it, a Wolf Head icon will appear on the map. And since its not mentioned in the other solutions, this Shrine can not be accessed until DNA Sequence 7. You can find the Shrine before this, but if you try to enter it, "Memory unavailable at this time" appears.
So you have to wait till DNA Sequence 7 to gain access to it. Also, you can reach the Shrine either by climbing down the ruins and jumping over to a ledge as shown in the video provided by Commander Drizzle. Or you can repair all 4 of the Broken Aqueducts in the Campagna District, which will fill the hole with water up to the entrance, making it easier to access.

Answered How do I unlock the last lair of romulus? You soon come to a large well that you need to climb down carefully. Jump from the wooden plank at the top to the opposite side of the well and tap the grab button to drop down. Tap the grab button while falling to grab a ledge. When you get near the bottom, slowly climb down to a landing from which you can turn around and jump to the next area. Continue following torches to get through the mazes.
When you reach a massive hall with a few followers of Romulus that scamper away, climb up the wooden structure on your left. You need to follow a series of jumps and hand holds around the eastern perimeter of the hall before turning west toward a giant cross. Climb the cross and jump west off the top of it to continue. Finally, you'll enter a circular room with a group of enemies. Fight off the crew on the bottom row and then look for a ladder that'll bring you to the second level with more enemies.
You need to hop from the outer walkway onto the center column of platforms, jumping back and forth between them to reach the top. You'll encounter enemies along the way but if you're trying to beat this stage quickly you can do your best to ignore them and continue to the end of the lair. I found this area particularly annoying, as you can fall if the enemies throw stones at you. Undertaker 2. Full Sync Requirement: Do not lose more than a block of health. The entrance to the lair is in the middle of a village, at the base of a wall on the inside of a walled-off courtyard.
Climb over the buildings to get to it. Start by running west through a doorway. The hall will eventually bring you to the upper level of the building, from which you can climb over a broken armoire to jump across the hall you started in, but on top of chandeliers.
Be careful not to fall from up high. To get full sync in this mission, you can't lose more than one block of health. There are no enemies to fight, but falling from significant distance will basically end your ability to get full sync. When you reach an area with a statue of a woman with her hands out, climb up onto the statue and jump left onto a hanging lantern.
As Ezio is flung off the lantern, you need to manually tap the grab button to latch onto some wood surrounding a column. From there you can shimmy around and continue. You'll swing down to a lower level before climbing back to the upper level and heading west. In a large room with a floor that gives out as you enter, hug the left south wall and use a thin, rickety wooden structure attached to the wall to grab a ledge ahead.
Soon you'll enter the final area, a massive Cathedral hall. Run to the north end and interact with a glowing gate. You need to equip a ranged weapon to hit a hanging sand bag left of the gate. If you have no ammo, search the area for containers on desks that will replenish your supply. Shooting the first sand bag opens up a passage to the upper level of the hall. You'll need to make your way to the northwest corner of the upper level before crossing back to the southeast corner.
There's a second sand bag to shoot down, causing another reaction. Use a leap of faith into a straw pile below and use the same passage you opened previously to get back to the second level of the hall. There's a third sandbag in the southwest corner to shoot down. This finally opens up the gate to the north, letting you grab the key from the main treasure chest and climb a ladder down to the lair's exit.
You can easily spot the entrance for this lair on the north side of a small stairway. Once underground, proceed until you wander into the middle of a mass that's ending.
Use Eagle Vision to spot the target Cardinal to the southwest. There are guards whose detection you need to avoid, and as well you need to stay hidden from the Cardinal. Blend in with a nearby group and follow them eastward.
The Cardinal will wander around a bit before moving through a gate to the south. Stay with him, but stay away from guards. Once through the gate yourself, it's time to chase down the Cardinal. The chase sequence makes up most of the mission and everything is pretty straight forward. Be quick, but don't rush things. Be methodical in your camera positioning so that all of your movements are straight and crisp, as making an error will waste more time than cautious progression.
Eventually you'll run the Cardinal into a dead end and he'll have to fight you. He doesn't put up much fight, making this quite easy. When he's dead, take a leap of faith out the window to grab the chest item and exit the lair. Getting to the entrance of this lair is a bit tricky.
The icon on the map leads you to the middle of a camp.
